Tamoxifen is metabolized in the liver. 
    In medicine, Tamoxifen is effective in the treatment of: breast cancer in postmenopausal women, breast cancer in men after castration, kidney cancer, melanoma (containing estrogen receptors), ovarian cancer, prostate cancer with resistance to other drugs.
    Male bodybuilders find this drug very effective in combating the effects of steroid aromatization and increased levels of estrogen in the body, such as gynecomastia and the formation of fat deposition, which forming feminine silhouette. The drug is not toxic and it fits well into most anabolic cycles.
    Tamoxifen if forbidden in case of: thrombophlebitis, pregnancy, increased sensitivity to tamoxifen, to drink alcoholic liquids in order to swallow tablets of this product.

Dosage:
    Dosing regimen is set individually, depending on the evidence, on patient's condition and on the scheme used as anticancer therapy. Tamoxifen can be administrate with food or without, at the same time each day. This enhances the effectiveness of Tamoxifen. You must swallow entire pill, without chewing it. Usually female who suffer of breast cancer, take Tamoxifen since 2 until 5 years. An average dose of Tamoxifen varies from 10 mg to 30 mg a day.
By athletes is used like 1-2tabs/day.

Drug interactions with Tamoxifen and its effect:
-In an application with anticoagulant coumarin derivatives, increased risk of higher anticoagulant action, 
-with cytostatic drugs - may increase the risk of thrombotic events.
-with allopurinol-possible hepatotoxicity, 
-with aminoglutethimide - decrease in plasma concentrations of tamoxifen, appears to be due to increasing its metabolism.
-Patients receiving tamoxifen may be noticed prolongation of neuromuscular blockade, induced by atracurium.
-With simultaneous use of bromocriptine may increase dopaminergic activity of bromocriptine.
-Patients receiving tamoxifen with warfarin, are at the risk of threatening the clinical situation: possible prolongation of the prothrombin time, hematuria, hematoma.
-While the application of mitomycin increases the risk of hemolytic-uremic syndrome.
-Concentration of Tamoxifen may be reduced in the blood plasma, which appears to be due to induction of CYP3A4 isoenzyme, under the influence of rifampicin.
-Estrogens may decrease the therapeutic effect of tamoxifen.

Side effects of Tamoxifen:
From the digestive system: nausea, vomiting, elevated liver enzymes, and in some cases - fatty liver, cholestasis, hepatitis.
CNS: rarely - depression, dizziness, headache, optic neuritis.
Eye disorders: rarely - retinopathy, keratopathy, cataract.
Hematopoietic system: rarely - thrombocytopenia, leukopenia.
Endocrine system: women - endometrial hyperplasia, vaginal bleeding, hot flashes, weight gain, men - impotence, decreased libido.
Cardio-vascular system: oedema, thrombosis, phlebitis.
Dermatological reactions: alopecia, rash, itching.
Other: Bone pain and lesions, fever.

Attention!
1. Should be carefully administrate in: leukopenia, thrombocytopenia, hypercalcaemia, patients with cataracts, hyperlipidemia.
2. During treatment should regularly monitor the peripheral blood picture (especially platelets), serum calcium and glucose levels; with prolonged use is advised ophthalmologist observation (every 3 months).
3. Should not be combined Tamoxifen with drugs, containing hormones, especially estrogen.
4. With the simultaneous use of Tamoxifen with drugs that affect the clotting of blood, needs a correction dose of tamoxifen.
5. In experimental studies found carcinogenic effect of tamoxifen.

Pregnancy and lactation:
    Tamoxifen is contraindicated during pregnancy. If necessary to use drug during lactation period, should be stop breastfeeding.

Overdose of Tamoxifen:
    Don't overdose this preparation. It may be dangerous. If you overdosed it, call your physician as soon as possible.

Excretion:
Excreted with the bile as metabolites.
